Description | Breeding for resistance (R) genes remains one of the most direct method of generating resistance in plants against pests and pathogens. Most R genes that have been characterized belong to the nucleotide-binding-site-leucine-rich-repeat (NLR) receptor family. Many NLR genes are known to play a role in resistance to pathogens. Their role in resistance to insects is relatively understudied. One example is the NLR gene named VAT that was reported to confer resistance to the aphid Aphis gossypii in melon (Cucumis melo). The characterization of NLRs in Cucurbita (pumpkin and squash) species has been done using available genomes. In this study, we aim to investigate the diversity of NLRs in three Cucurbita species (C. maxima, C. pepo, and C. moschata), and study their potential role in aphid resistance. Therefore, in this project you will use several accessions that have been classified as resistant or susceptible to the aphid A. gossypii. You will also determine the presence/absence of VAT or VAT related genes in the selected accessions. In addition, this will contribute towards the discovery of novel R-genes against pests in Cucurbita species. |
Used skills | Molecular techniques and bioinformatics such as DNA extraction, primer design, PCR, use of sequence analysis tools such as BLAST and multiple sequence alignment software, and understand plant genetics. Ability to analyze and interpret the data and write a thesis. |
